Objectives. To evaluate the sperm chromatin dispersion in men with infertility using the first Russian kit.
Methods. The study was conducted between July and August 2022. Study participants were 492 male volunteers aged 23 to 59 years with infertility which semen samples were examined by GEMSTANDART–HaloSperm L&Q kit so-called
Halo test. The outcomes were as a percentage as the sperm DNA Fragmentation Index (DFI), %. The results of Halo test were evaluated according to the following threshold values: in reference range (normal): DFI ? 15 %, controversial result: from DFI > 15 % to < 25 %, out of reference range (pathological) DFI > 25 %. All of men (n = 492) had the results of a basic semen examination which performed a month before the current clinical trials with a various scenario of pathospermia as well as normospermia. The statistical processed a GraphPad Prism software (version 8.0.1) and Excel statistical package Microsoft? Excel? for Microsoft 365 MSO (verse 2201 16.0.14827.20158). The Pearson’s rank correlation coefficient of parametric measure and descriptive statistics have been applied.
Results. Sperm DNA fragmentation (SDF) was not detected in 54.1 % (266/492 = 54.1 %) men whose average age was 36.58 years (DFI ? 15 %); a poor direct correlation with normal level of DFI and asthenozoospermia (r = 0.20; p < 0.0001), viscosipathy (r = 0.13; p < 0.0001) are presented; a poor inverse correlation with oligoasthenoteratozoospermia (r = – 0.12; p < 0.0001) are presented; the absence of leucospermia was noted.
A controversial results for SDF presence was found in 21.3 % (105/492 = 21.3 %) men whose average age was 38.71 years (DFI range from > 15 to < 25 %); a poor direct correlation with oligoasthenoteratozoospermia (r = 0.24; p < 0.0001), oligospermia (r = 0.23; p < 0.0001) and average age of 39 years (r = 0.17; p < 0.0001) was noted; poor inverse correlation with asthenozoospermia (r = – 0.22; p < 0.0001) was presented.
SDF was found in 24.6 % (121/492 = 24.6 %) men whose average age was 41.55 (DFI > 25 %); poor direct correlation with a high level of DFI and average age for infertile men in 44 years (r = 0.28; p < 0.0001) was presented.
All semen samples (n = 492) presented a poor positive correlation with DFI and age (r = 0.1563; p = 0.0005); oliasthenoterato-zoospermia (r = 0.19; p = 0.0001), leucospemia (r = 0.29; p = 0.0001), and necrozoospermia (r = 0.16; p = 0.0004).
Conclusion. The results of the study showed that GEMSTANDART–HaloSperm L&Q kit complements a basic semen examination providing specific information to the clinicians about quality of genetic material in semen samples.
Objectives. The aim of the study was to estimate the protection level by anti-IgG to RBD S-protein of the SARS-CoV-2 delivered from natural and vaccine-induced immunity in persons who have contact with COVID-19.
Methods. Fourteen (14) persons presented a lab medical stuff and their relatives aged 19-72 years with several COVID-status (CoV-ST) who had contact with COVID-19 the last 10 days before blood collection was underwent to clinical trials.
The CoV-ST presented by 1) non-vaccinated, recovered, nV-Recover; 2) unvaccinated, non-contact, V-nC; 3) recovered and then vaccinated (hybrid), R-V (hybrid); 4) vaccinated and then infected and recovered, V-R; 5) vaccinated, now ill, |V-Ill; 6) non-vaccinated, now ill, nV-Ill. Quantitative ELISA was conducted by anti-IgG RBD S-protein SARS-CoV-2 kit in blood serum with resulting in BAU “binding antibody units” (First WHO International Standart for anti-SARS-CoV immunoglobulin (human), NIBSC code: 20/136). The estimation was performed according manual for kit and immunoassay analyser in optical density of solutions at one main wavelength of 405 nm. The Spearman’s rank correlation coefficient of nonparametric measure was used due to small numerous of cohort. Statistical analyses was calculated with GraphPad Prism 8.0.1 and Microsoft? Excel? for Microsoft 365 MSO (verse 2201 16.0.14827.20158) programs.
Results. All fourteen (14) persons who had contact with COVID-19 was immunized. Three (3) female persons aged less than 30 years had anti-IgG RBD S-protein SARS-CoV-2 in diapason from 19.2 till 230.1 BAU/mL which was infected and got ill with might-to-moderate symptoms of COVID-19 (f,34 nV-Recover; f,33 nV-Ill; f,22 V-Ill). Eleven (11) persons which was not infected and got ill 1) six (6) with R-V (hybrid) and vector technology vaccine had anti-IgG < 500 BAU/ml; 2) eight (8) persons, seven (7) of them with R-V (hybrid), vector and mRNA-technology vaccines had anti-IgG >500 BAU/ml; 3) four (4) persons with R-V (hybrid), one (1) of them had a vector technology vaccine, three (3) of them had an mRNA technology vaccine had anti-IgG > 5000 BAU/ml; 4) the highest concentration of specific anti-IgG 50,000 BAU/ml was detected in 19-year-old men with R-V (hybrid) and mRNA technology vaccine.
Conclusion. Direct moderate and moderate inverse correlation was noted between many factors which can determine an adaptive human immunity. The observe study showed a rapid immune response to neutralization of viruses in medical stuff and their relatives who had closest contact with COVID-19, aged 19 to 50 years and who previously was ill and subsequently was vaccinated triply by vector vaccine or previously was ill and then was vaccinated twice by mRNA vaccine.
Objective: to estimate the effect of ejaculate consistency on the levels of interleukin-6 and interleukin-8 in human seminal plasma.
Material and methods. The concentration of interleukin-6 and interleukin-8 was determined by ELISA using the kit manufactured by ?Vector-Best? (Russia). The study included 64 men: the main group (n = 30) presents patients with high semen viscosity, the comparison group (n = 34) presents men with normal semen viscosity.
Results. In average, interleukin-6 level in the semen was 13.45 pg/ml, the median was 13.79 pg/ml; the data ranged from 8.24 pg/ml to 19.34 pg/ml. In average, level of interleukin-8 was 28.9 pg/ml, the median – 13.96 pg/ml; there is a large range of values from 0.202 pg/ml to 174.5 pg/ml. There are no significant differences in the values of interleukin-6 and interleukin-8 of the main group from the comparison group: for interleukin-6, U = 377.0 (p = 0.074655); for interleukin-8, U = 407.0 (p = 0.863852). The data obtained did not correlate neither between groups nor with the fertility markers of the human semen.
Conclusion. Interleukin-6 and interleukin-8 levels in the human seminal plasma do not depend on semen viscosity.
This article discusses the possibilities of application of artificial neural networks to solve problems of increasing the diagnostic outcomes in clinical laboratory examination. High diagnostic sensitivity (96 %) and diagnostic accuracy (89.5 %) of the results were shown on a large amount of cellular material digitized by artificial intelligence microscopy automation system like the Vision Cyto Pap.
The high resolution and sharpness of digital slides, the mode of viewing objects (cells) in the gallery, quick access to the results of preclassification, all of these factors together allow to reduce turnearound time in more than 2.5 times reducing disadvantages of the microscopy.
Application of artificial neural networks does not substitute a doctor’s skills. The role in validation of reports eligible only for cytopathologist. This concept indicates a carefully approach for staff working with a microscope, respectful attitude to them professional skills, and highlights a personalized approach to patients..
